1. Project Background
Myoglobin (Myo) is a binding protein comprised of a peptide chain and a heme prosthetic group. This protein plays a vital role in storing oxygen in muscles. Elevations in myoglobin levels can occur as early as 2 hours after the onset of chest pain. Patients with severe congestive heart failure and those who undergo cardiac surgery may also experience increased levels due to myocardial damage. Given its sensitivity, myoglobin serves as a crucial indicator for diagnosing acute myocardial infarction, thus establishing itself as one of the current markers for myocardial infarction.
2.Performance
2.1 Chemiluminescence (acridinium ester) platform:
Combination: Human myoglobin (Myo) antibody (Cat. No.: EKY0087Q) coated carboxyl magnetic beads + human myoglobin (Myo) antibody (Cat. No.: EKY0087R) labeled acridinium ester;
Linear range: 5ng/mL-1000ng/mL; anti-HOOK concentration can reach 30000ng/mL.
2.2 Fluorescence immunochromatography (lanthanide Eu chelate filling) platform:
Combination: Human myoglobin (Myo) antibody (Cat. No.: EKY0087Q) labeled fluorescent microspheres + human myoglobin (Myo) antibody (Cat. No.: EKY0087R) for streaking;
Linear range: 5ng/mL-1000ng/mL.
3. Clinical relevance
3.1 Chemiluminescence (acridinium ester) platform:
Clinical correlation with R brand Myo reagent R2>0.98
The correlation between the low value part (≤300ng/mL) and R brand Myo reagent is R2>0.96.
3.2 Fluorescence immunochromatography (lanthanide Eu chelate filling) platform:
Clinical correlation with R brand Myo reagent R2>0.98;
The correlation between the low value part (≤300ng/mL) and R brand Myo reagent is R2>0.95.
4. Test plan
4.1 Chemiluminescence (acridinium ester) platform:
Antigen to be tested: Human Myoglobin protein (Cat. No.: EKY0105A), sample/standard: adding volume 10ul;
Reagent 1: Human myoglobin (Myo) antibody (Cat. No.: EKY0087Q), coated-carboxy magnetic bead dilution, dosage 50ul/test;
Reagent 2: Human myoglobin (Myo) antibody (Cat. No.: EKY0087R), labeled-acridinate dilution, dosage 50ul/test;
Reaction conditions: incubation temperature 37°C, sample + reagent 1 + reagent 2 (10 min);
Luminescence instrument used in the test: Cosmai SMART 500S; chemiluminescence instrument that can be used for various acridinium ester systems (such as Sichuan Mike, Shenzhen Yahuilong, Shandong Kanghua Bio, Getein Bio, Nanjing Norman, etc.)
4.2 Fluorescence immunochromatography (lanthanide Eu chelate filling) platform:
Antigen to be tested: Human Myoglobin protein (Cat. No.: EKY0105A), sample/standard: adding volume: 5ul;
Reaction conditions: mix sample + fluorescent solution, load sample, reaction time (10min-15min);
Fluorescence instrument used for testing: Hemai FIC-S100; instruments that can be used in various fluorescence systems (excitation wavelength 365nm, emission wavelength 615nm) (such as Guangzhou Labsim, etc.)
