Choosing the Right Surfactant: A Simple Guide

Surfactants are substances that can create a film at interfaces, form micelles in solvents, and significantly reduce the surface energy of water.
Surfactants hold an amphoteric molecular structure, meaning they have two ends: a hydrophilic group, also known as oleophobic, which allows the surfactant to dissolve in water in a monomeric form; and a hydrophobic group, also known as lipophilic, which is typically a non-polar hydrocarbon chain.
Hydrophilic groups are often polar, like carboxyl (-COOH), sulfonate (-SO3H), amino (-NH2) or amine groups and their salts, hydroxyl (-OH), amide groups, ether bonds (-O-), etc. On the other hand, hydrophobic groups are usually non-polar hydrocarbon chains, like hydrophobic alkyl chains (R-), Ar- (aryl), etc.
Surfactants are categorized into ionic surfactants (including cationic and anionic), nonionic surfactants, amphoteric surfactants, compound surfactants, and others.
In a surfactant solution, when the concentration reaches a certain value, the molecules will form micelles. Micellarization, or the creation of micelles, is a fundamental property of surfactant solutions, with important interfacial phenomena linked to their formation. The concentration at which surfactants form micelles is termed critical micelle concentration (CMC). Micelles are not fixed in shape but have dynamic and irregular structures. Under specific conditions, surfactants can also form reverse micelles.
Several factors affect critical micelle concentration:
- The structure of the surfactant
- The addition and type of additives
- The impact of temperature
Protein Interactions with Surfactants:
Ionic surfactants primarily engage in electrostatic interactions with proteins, where polar groups interact with charged regions, and hydrophobic hydrocarbon chains interact with the hydrophobic parts of proteins. This leads to the formation of a P-S complex. Non-ionic surfactants predominantly interact with proteins through hydrophobic forces. The interaction between their hydrophobic chains and the hydrophobic groups of proteins can impact the structure and function of both surfactants and proteins. Hence, the surfactant's type, concentration, and system environment determine whether it stabilizes or destabilizes proteins, causing aggregation or dispersion.
HLB Value:
For surfactants to exhibit effective interfacial activity, there must be a balance between hydrophobic and hydrophilic groups. The Hydrophile-Lipophile Balance (HLB) is a measure indicating this balance and serves as a guide for selecting surfactants.
The HLB value, ranging from 0 to 40, is a relative indicator of a surfactant's hydrophilic and hydrophobic properties. For instance, paraffin has an HLB value of 0 (no hydrophilic group), polyoxyethylene is 20, and SDS, known for its strong hydrophilicity, has an HLB value of 40. A larger HLB value indicates better hydrophilicity, while a smaller value indicates poorer hydrophilicity.
